Are the one syllable and two syllable “blessed” defined the same?
 
Depends (at least in my opinion).

1 syllable = verb As in “The baby was blessed by a priest.”

2 syllables = adjective As in “Have a blessed (bless-ed) Christmas.”

Don’t know the official defination though.
 
When it is used as the past tense of the verb “bless”, it has one syllable. When used as a description or a title (i.e. Blessed Mother Theresa), it has two syllables.

Blessed be God?

If we are blessed by God, how, as creatures,
are we allowed to bless Him in return?

To bless is to show favor toward something or someone. When we say God has blessed us, it means He has shown His mercy and love to us in some way. When we say that we bless God, it means that we praise and honor Him. Does that help?
