Is the prenuptial investigation prior to a marriage only to establish whether a couple is free to marry, or does it also have an effect on the validity of the marriage? Take, for example, a couple where one of the spouses had a previous marriage that had been annulled, but where the spouse in question lied to the priest in the investigation about the existence of this previous marriage. Would the couple still validly marry because in reality they are free to marry, or would a lie during the investigation render their attempt at marriage invalid?

No, the prenuptial investigation does not affect the validity of a marriage so long as both parties are free to marry and have the intention to enter marriage as the Church understands it.
The prenuptial investigation is to establish those facts. I have no idea why someone would lie about being previously married with an annulment. While the person is objectively free to marry and lying about that wouldn’t affect the validity of the marriage, the motivations for lying might. Is this individual trying to deceive their fiance? Obtaining consent from one’s spouse through deceit could invalidate a marriage. But lying to the priest would not invalidate due to the objective reality necessary for a marriage being properly in place.
