Now take a look at these two parallel verses from the KJV.
So a question is being asked here. Also an ASSUMPTION is being made as to the relatives of Jesus.
They WRONGLY assume Jesus is the son of Joseph(the carpenter) because they did not know the TRUTH of his parentage at this moment in time. IE They did not know that Jesus was not actually the son of Joseph, but ACTUALLY was the Son of God. So the same Assumption is being applied to these Brothers and Sisters. They could just be the offspring of Joseph.
I remember my brother critiquing “The Passion” by Mel Gibson. He asked “Where is Joseph”. I replied "Where is here in the Bible after Jesus was taken to the Temple in Luk_2:27 as a child? As a Protestant Pastor, he had no answer for that.
Tradition holds that Joseph was elderly and that his Betrothal to Mary was only “Custodial” in nature.
And you will notice that in one verse Brethren is used and in the other uses Brother. Both come from the same Greek Word which:
Both are recording the same event.Mat_13:55 Is not this the carpenter’s son? is not his mother called Mary? and his brethren, James, and Joses, and Simon, and Judas?
Mar_6:3 Is not this the carpenter, the son of Mary, the brother of James, and Joses, and of Juda, and Simon? and are not his sisters here with us? And they were offended at him.

And the Greek word for Sister in Mark’s:From G80; brotherhood (properly the feeling of brotherliness), that is, (the (Christian) fraternity: - brethren, brotherhood
